Progress Report: PHGDH ACT Domain — Lead Optimization for CNS-Penetrant AD Therapeutic (Cycle 1)
Progress Report: PHGDH ACT Domain — Lead Optimization for CNS-Penetrant AD Therapeutic (Cycle 1)
Executive Summary
This is the first cycle of the research project. No previous data is available for comparison.
Research Progress
In this cycle, we established the structural foundation for the lead optimization campaign. The primary PDB structure (6RIH) was substituted with the high-confidence AlphaFold model (AF-O43175-F1), which provided 100% coverage of the target ACT domain residues (395-480). The binding site preparation tool successfully extracted the pocket geometry, yielding coordinates (center: 17.8, 19.21, -21.1).

NCT-503 ranked #1 with a total score of 0.96, primarily due to its high rbd_mechanistic_evidence score of 5. The top 3 compounds were NCT-503, BI-4924, and CHEMBL3675086, with total scores of 0.96, 0.93, and 0.68, respectively. A surprising finding was the low ranking of CHEMBL3656205, despite its high synthesizability score of 5.
